Slant cryopreservation method This is the simplest preservation method that puts the growth-free, pollution-free beveled tube into the refrigerator, preserved at a low temperature of 4°C to 6°C, and then transferred to fresh water every 2 to 3 months. The slant medium, after cultivation, continue to preserve (to supplement the nutrients and moisture of mycelium, to prevent the aging of hyphae).
Liquid paraffin preservation method This method is to inject the paraquat in the aseptic chamber in the beveled tube to be preserved, and to inject the wax to 1 cm above the slope. Its purpose is to isolate bacteria from the air and reduce its metabolic activity. Then stuffed with a cork or rubber stopper, sealed with wax, upright placed in a dry place indoors, can be stored for more than one year.
When using, only a small amount of mycelium is picked from the slant with an inoculation needle and transplanted on a fresh slant medium. Since the hyphae are stained with paraffin, it must be transferred to a test tube to grow well.
